Detection and mapping of a lethal locus in a eucalyptus hybrid population PAB
Rosado,Tatiana Barbosa; Tomaz,Rafael Simões; Rocha,Rodrigo Barros; Rosado,Antônio Marcos; Alves,Alexandre Alonso; Araújo,Elza Fernandes de; Alfenas,Acelino Couto; Cruz,Cosme Damião.
The objective of this work was to verify the existence of a lethal locus in a eucalyptus hybrid population, and to quantify the segregation distortion in the linkage group 3 of the Eucalyptus genome. A E. grandis x E. urophylla hybrid population, which segregates for rust resistance, was genotyped with 19 microsatellite markers belonging to linkage group 3 of the Eucalyptus genome. To quantify the segregation distortion, maximum likelihood (ML) models, specific to outbreeding populations, were used. These models consider the observed marker genotypes and the lethal locus viability as parameters. The ML solutions were obtained using the expectation‑maximization algorithm. A lethal locus in the linkage group 3 was verified and mapped, with high confidence,...

Genetic control of Eucalyptus urophylla and E. grandis resistance to canker caused by Chrysoporthe cubensis Genet. Mol. Biol.
Guimarães,Lúcio Mauro da Silva; Resende,Marcos Deon Vilela de; Lau,Douglas; Rosse,Leonardo Novaes; Alves,Alexandre Alonso; Alfenas,Acelino Couto.
Chrysophorte cubensis induced canker occurs in nearly all tropical and subtropical regions where eucalypts are planted, causing losses in both wood quality and volume productivity, especially so in the warmer and more humid regions of Brazil. The wide inter and intra-specific genetic variability of resistance to canker among Eucalyptus species facilitates the selection of resistant plants. In this study, we evaluated resistance to this pathogen in five Eucalyptus grandis (G) and 15 E. urophylla (U) trees, as well as in 495 individuals from 27 progenies derived from crosses between the trees. In the field, six-months-old test seedlings were inoculated with C. cubensis. Lesion length in the xylem and bark was measured eight months later. The results...

Isozymic variability in a Brazilian collection of annatto (Bixa orellana L.) PAB
Carvalho,Jane Fiuza Rodrigues Portela de; Robinson,Ingrid Peters; Alfenas,Acelino Couto.
The objectives of this work were to optimize the isozyme electrophoresis technique for Bixa orellana, and use isozyme markers for a preliminary survey on the genetic variability in Brazilian annatto germplasm accessions. Collection consisted of seed samples from sixty open pollinated trees, representing two Northern and four Southern geographic provenances. The extraction, electrophoresis, and interpretation of annatto isozymes are described. Three out of the twenty-one identified isozyme loci were polymorphic in the collection. The percentage of polymorphic loci (P = 21.05) and the expected heterozygosity in annatto (H T = 0.064) were low, compared to other tropical woody species. A UPGMA phenogram, constructed with Nei's genetic distances, clearly...

Linkage analysis between dominant and co-dominant makers in full-sib families of out-breeding species Genet. Mol. Biol.
Alves,Alexandre Alonso; Bhering,Leonardo Lopes; Cruz,Cosme Damião; Alfenas,Acelino Couto.
As high-throughput genomic tools, such as the DNA microarray platform, have lead to the development of novel genotyping procedures, such as Diversity Arrays Technology (DArT) and Single Nucleotide Polymorphisms (SNPs), it is likely that, in the future, high density linkage maps will be constructed from both dominant and co-dominant markers. Recently, a strictly genetic approach was described for estimating recombination frequency (r) between co-dominant markers in full-sib families. The complete set of maximum likelihood estimators for r in full-sib families was almost obtained, but unfortunately, one particular configuration involving dominant markers, segregating in a 3:1 ratio and co-dominant markers, was not considered. Here we add nine further...

Rhizobacterial characterization for quality control of eucalyptus biogrowth promoter products BJM
Zarpelon,Talyta Galafassi; Guimarães,Lúcio Mauro da Silva; Alfenas-Zerbini,Poliane; Lopes,Eli Sidney; Mafia,Reginaldo Gonçalves; Alfenas,Acelino Couto.
Abstract Plant growth-promoting rhizobacteria strains from special formulations have been used to optimize eucalyptus cutting production. To undertake quality control for the formulated products, the rhizobacterial strains should be characterized to assess their purity and authentication. In the present study, we characterized nine strains of rhizobacteria, including three Bacillus subtilis (S1, S2 and 3918), two Pseudomonas sp. (MF4 and FL2), P. putida (MF2), P. fulva (Ca), Frateuria aurantia (R1), and Stenotrophomonas maltophilia (CIIb). The strains were differentiated by colony morphology after 24 h of incubation in three different solid state culture media (glucose-nutritive agar, 523 medium and yeast extract-mannitol agar), sensitivity to a panel of...

Rhizobacterial promotion of eucalypt rooting and growth BJM
Teixeira,Débora A.; Alfenas,Acelino Couto; Mafia,Reginaldo Gonçalves; Ferreira,Eraclides M.; Siqueira,Leandro de; Maffia,Luiz A.; Mounteer,Ann H..
A total of 107 rhizobacterial isolates, obtained from the rhizosphere of eucalypt clones were tested as rooting inducers of cuttings and mini-cuttings planted in substrate composed of carbonized rice husk and vermiculite (1:1). Cuttings and mini-cuttings were planted in conical plastic tubes containing treated and untreated (control) substrate and kept under intermittent mist irrigation at 26-28ºC. After 35 days, rooting percentage and dry root matter of cuttings were evaluated. Ten isolates capable of providing gains of up to 110% in root formation and up to 250% in root biomass over non-inoculated control cuttings were selected. Gains in rooting varied according to clone and isolate tested. The greatest gains were obtained for the mini-cuttings...

The use of self-incompatibility in the production of hybrid eucalyptus seed by `Aracruz Celulose' in Brazil Genet. Mol. Biol.
Junghans,Tatiana Góes; Peters-Robinson,Ingrid; Bertolucci,Fernando L.; Alfenas,Acelino Couto.
Self-incompatibility found in a Eucalyptus grandis clone was used to promote interspecific hybridization between E. grandis and E. urophylla clones. The interspecific hybridization achieved in an open-pollinated commercial seed orchard planted in Espírito Santo, Brazil was evaluated by the multi-locus outcrossing rate (tm) of the seed producing clone, E. grandis. The percentage of outcrossed seeds reached 95.9%. The outcrossing rate of individual trees was quite variable, but was always above 70.0%. Wright's coefficient was negative (F = -0.30) revealing an excess of heterozygous genotypes in the progeny. Genetic parameters confirmed the high degree of hybridization expected in this orchard. The 800-m wide protection belt of native tropical forest that...
